Ipconazole Induces Oxidative Stress, Cell Death, and Proinflammation in SH-SY5Y Cells

Ipconazole is an antifungal agrochemical widely used in agriculture against seed diseases of rice, vegetables, and other crops; due to its easy accumulation the environment, it poses a hazard human, animal, environmental health. Therefore, we investigated cytotoxic effect ipconazole on SH-SY5Y neuroblastoma cells using cell viability tests (MTT), ROS production, caspase3/7 activity, molecular assays biomarkers death (Bax, Casp3, APAF1, BNIP3, Bcl2); inflammasome (NLRP3, Casp1, IL1β); inflammation (NFκB, TNFα, IL6); antioxidants (NRF2, SOD, GPx). were exposed (1, 5, 10, 20, 50, 100 µM) for 24 h. The ipconazole, dose-dependent manner, reduced produced IC50 32.3 µM; also increase production enzyme activity cells. In addition, at 50 µM induced overexpression Bax, BNIP3 (cell genes); NLRP3, IL1B (inflammasome complex NFκB, IL6 (inflammation expression NRF2, GPx (antioxidant genes). Our results show that produces effects by reducing viability, generating oxidative stress, inducing cells, so exposure should be considered as factor presentation neurotoxicity or neurodegeneration.
